A cell/tissue culture premising the transplant is the process that is important from the viewpoint of production, and the process and quality control for safety and the efficacy is indispensable in order to supply a final cell/tissue product with uniform and stable quality. As for evaluation technologies, those concerning safety have been constructed considerably up to now, but those concerning efficacy is still insufficient.

In this lecture, the planar culture design enabling cell morphology control, the automation of cultivation process, and the stereological evaluation of the cultured tissues for the chondrocyte, myoblast and mesenchymal stem cell will be presented widely from the culture process control to the quality control. The contribution of chemical engineering to these technology developments will also be discussed.

The articular cartilage reproduction by the cell transplant is one of the treatment methods that have been clinically applied since the earliest time. The autologous chondrocytes transplant was reported in rabbits in 1989, and became the tissue regeneration technological method that FDA authorized first in the orthopedics field of the world in 1997. Since then the treatment has been executed in more than 20,000 cases all over the world. However, its availability is being still controverted.

At first, Wakitani and his colleagues had made an attempt on allogeneic chondrocytes transplant, but they abandoned its approach because of the difficulty for cell collection in Japan. Afterwards, they started the research on the bone marrow mesenchymal cell (BMMC) transplantation, and tried the clinical application in advance of the world in 1998, but it has not come to be generalized yet. In this lecture, their recent research results will be presented together with their efforts for the generalization (industrialization).

The cancer stem cell plays an important role to generation, the treatment resistance, the recurrence, and the metastasis, etc. of the malignant tumor. The cancer stem cell research with the possibility of connecting directly with the clinical application has been preceded in the hematopoietic organ tumor, and currently the verification is advanced aggressively also as for a solid tumor. In this seminar, the trend of the cancer stem cell research on a solid tumor will be summarized first, followed by presentations of updated research results leading to the clinical application in the near future, which help young researchers to improve their skills.

The cell transplantation is extremely promising as a new treatment for cardiac failure, and its clinical application is pushed forward in each country.However, further basic studies in various respects are necessary and indispensable so that this treatment approach may succeed in the future.
Suzuki and his colleagues, in order to develop more safe and effective strategy in this approach, have been clarifying dynamic state and changes of transplanted cell and its direct/indirect effects on the cardiac muscle from various points of view.
In this lecture, their recent works, especially on paracrine effects caused by the cell transplantation, will be presented in detail.

The 45th MTR Seminar " Regeneration therapy based on underlying mechanism of heart failure" by Prof. Issei Komuro, Department of Cardiovascular Science and Medicine, Chiba University Graduate School of Medicine.

  Cardiac hypertrophy occurs as a compensatory response to various stresses. However, collapse of the compensation causes heart failure. Prof. Komuro and colleagues have clarified that the cardiac angiogenesis is crucially involved in the adaptive mechanism of cardiac hypertrophy and that an accumulation of tumor suppressor gene p53 is essential for the transition from cardiac hypertrophy to the heart failure (Sano, et al. Nature 2007)

  They treated about 80 patients with peripheral arterial obstruction by angiogenesis therapy using peripheral blood mononuclear cells, and found effectiveness in over 70 percent of them. They are planning to apply the therapy to patients with cardiac failure.

  In addition, they identified IGFBP-4 as a potent enhancer of cardiomyocyte differentiation (Zhu, et al., Nature, 2008). IGFBP-4 played a role indispensable to cardiomyocyte differentiation and even to cardiogenesis by down regulating wnt signal . The possibility of its promoting effect on cardiac regeneration is currently under investigation.

The Knowledge Cluster Initiative (started by The Ministry of Education, Culture, Sports, Science and Technology (MEXT) in 2007) is promoting "Drug Discovery", "Regenerative Medicine", and "Prevention of the Lifestyle Diseases". In this Symposium, research progress obtained so far will be presented.

Recently, emerging and reemerging infectious diseases such as avian influenza, SARS, BSE, and drug resistant bacteria infection have brought various influences to our country and the world. In this seminar the following presentations are scheduled:

2009-1-9 (Friday):
The Senri Life Science Seminar "Stem Cell and Pluripotency"


Prof. Yamanaka reported on the establishment of human iPS cell line on Nov. 2007. For this worldwide discovery Japanese government decided the research support with exceptional speed, and held the symposium of iPS at the end of last year. Subsequently, the research on iPS cell has accelerated from various angles all over the world. In fact, the session of iPS attracts the most a lot of audiences in any conference on the stem cell.

  This seminar is held in year about first from the Yamanaka's report, so it would be valuable for similar members of the JST Symposium 2007 including Prof. Yamanaka to look back on what has been elucidated in this one year.

  In this Seminar they will discuss freely about what can be done, what has not been realized, what kind of researches has been conducting in the world, what is the future as a result, and what is the problem to Japan generating this technology.

2008-12-10 (Wednesday):
The 44th MTR Seminar "Customization of Extracellular Matrix and Matrixome " by Dr.Kiyotoshi Sekiguchi, Professor of the Institute for Protein Research Osaka University.


The cell proliferation and differentiation are regulated by various information released from the surrounding environment. The players bearing such information are the extracellular matrix and humoral factors secreted from the neighboring cells.

The lecturer has focused on the basement membrane as a scaffold closely available for most cells, and comprehensively analyzed the localization of almost all known basement membrane proteins using the immunohistochemical method.

In this seminar, he will introduce the molecular entity of extracellular matrix clarified through his recent analysis, focusing especially on molecular composition of the basement membrane and its spatiotemporal control customized for each cell. In addition, he will discuss its significance in the regenerative medicine.

The Opening Ceremony of "Saito Bio-innovation Center" will be held according to the following. In the Saito Life Science Park, two incubation facilities have been operated for bio-ventures so far, and the third incubator has been anticipated because of their full occupancy state. All the facilities including "Saito Bio-innovation Center" have already become full, and 35 venture companies related to biotechnology are now advancing their research and development.

Special seminar "New development of stem cells derived from fat tissue" by Dr. Akifumi Matsuyama, Associate Professor of MTR (Medical Center for Translational Research, Osaka University Hospital)


NIBIO (National Institute of Biomedical Innovation) will invite Dr. Akifumi Matsuyama, Associate Professor of MTR (Medical Center for Translational Research, Osaka University Hospital), and hold a special seminar. Dr. Matsuyama found polyphyletic progenitor cells as new stem cells from fat tissue that can be harvested easily and safely. The new stem cells are characterized by expression of Isl-1, a non-differentiation marker, and have ability to differentiate not only into bone, cartilage, fat tissue, but also into cardiac myoblasts, hepatic cells, insulin-secreting cells. Additionally, these new stem cells partially express c-myc, oct3/4, Klf4 and nanog.
Please participate by all means since the application of such new stem cells are expected very much for development toward regenerative therapy and drug discovery screening.

The associate professor Collins specializes in the research of politics and the science & technology policy, and is currently doing a comparative study on the biotechnology cluster in the world from the policy side. He is especially well informed about the biotechnology cluster circumstances of the United States and Japan, and the main includes a comparison research on the biotechnology cluster of Kobe and its sister city Seattle where University of Washington locates, and a study on Research Triangle Park in North Carolina, etc.
Because he speaks Japanese a little, the chance of the question and answer is scheduled, though Nagao, associate professor of Osaka City University Graduate School Economics, interprets in the lecture.
